ABSTRACT

Objective By using the approach of "hierarchy three-level index system",this study aims at achieving precise scientific research performance evaluation, improving the proportion of high-tech products, and complementing the accumulative effect of conventional evaluation system. Methods This study transforms the conventional "plat three-level index system" into ‘hierarchy three-level index system',which consists of three levels of indexes, namely the national level, provincial level and local level.Candidates are categorized into three echelons according to the criteria for inclusion.Candidates of each echelon are evaluated and ranked by three-level indexes.Fuwai hospital's scientific advanced departments and individuals were evaluated by these two methods mentioned above, and the results were compared. Results These two index system are basically the same regarding to the overall evaluation pattern.Hierarchy three-level index system better displays the‘ quality strategy',allowing units with greater proportion of high-tech products to stand out as well as reducing the accumulative effect of massive production of medium-and-low-end products. Conclusions "Hierarchy three-level index system" is the remedy for the deficiencies of the conventional "plat three-level index system",it improves the reliability and validity of technology evaluation. Therefore, the system will contribute to the selection of scientific brains and the establishment of technology innovation system.

ABSTRACT

To explore the clinical features and common complications of fast resting heart rate (RHR) in hypertensionpatients. Methods: We retrospectively analyzed the entire rest electrocardiogram data of Qingdao study 2000 and Xinyang study2005 in community population elder than 18 years including hypertension patients and non-hypertension subjects. Clinical complications as diabetes, coronary artery disease, hyperlipidemia and stroke, laboratory findings, RHR in ECG, body mass index (BMI), waist to hip ratio and office blood pressure were collected in all participants. Results: A total of 18183 participants were enrolled including 61.6% male, the average age was (45.2±12.7) years including 6763 hypertension patients. Compared with normal BP subjects, hypertension patients had the faster RHR (73.5±11.6) times/min vs (70.6±9.6) times/min, P<0.001 and more hypertension patients combining fast RHR (14.5% vs 6.4%), P<0.001. In hypertension patients, compared with normal RHR patients, fast RHR patients had the elder age (53.9±12.2) years vs (51.8±11.2) years, lower BMI (25.8±3.6) kg/m2 vs (26.4±3.4) kg/m2 and higher ratio of grade 3 hypertension (68.2%vs 59.0%), all P<0.001; higher levels of fasting blood glucose (6.0±2.4) mmol/L vs (5.6±1.5) mmol/L and triglyceride (2.0±1.8) mmol/L vs (1.7±1.3) mmol/L, both P<0.001, higher LDL-C (3.2±0.9) mmol/L vs (3.1±0.8) mmol/L, P=0.001;more patients with diabetes (6.6% vs 3.9%), P=0.007 and stroke (11.1% vs 8.3%), P=0.005. Multivariate regression analysis indicated that with adjusted traditional risk factors, fast RHR was positively related to stroke occurrence in hypertension patients (OR=1.306, 95% CI 1.021-1.671). Conclusion: Fast RHR happened more in hypertension patients than in normal BP subjects; it had the increased risk for stroke occurrence in hypertension patients.

ABSTRACT

<p><b>BACKGROUND</b>Green tea has been shown to improve cholesterol metabolism in animal studies, but the molecular mechanisms underlying this function have not been fully understood. Long non-coding RNAs (lncRNAs) have recently emerged as a major class of regulatory molecules involved in a broad range of biological processes and complex diseases. Our aim was to identify important lncRNAs that might play an important role in contributing to the benefits of epigallocatechin-3-gallate (EGCG) on cholesterol metabolism.</p><p><b>METHODS</b>Microarrays was used to reveal the lncRNA and mRNA profiles in green tea polyphenol(-)-epigallocatechin gallate in cultured human liver (HepG2) hepatocytes treated with EGCG and bioinformatic analyses of the predicted target genes were performed to identify lncRNA-mRNA targeting relationships. RNA interference was used to investigate the role of lncRNAs in cholesterol metabolism.</p><p><b>RESULTS</b>The expression levels of 15 genes related to cholesterol metabolism and 285 lncRNAs were changed by EGCG treatment. Bioinformatic analysis found five matched lncRNA-mRNA pairs for five differentially expressed lncRNAs and four differentially expressed mRNA. In particular, the lncRNA AT102202 and its potential targets mRNA-3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) were identified. Using a real-time polymerase chain reaction technique, we confirmed that EGCG down-regulated mRNA expression level of the HMGCR and up-regulated expression of AT102202. After AT102202 knockdown in HepG2, we observed that the level of HMGCR expression was significantly increased relative to the scrambled small interfering RNA control (P < 0.05).</p><p><b>CONCLUSIONS</b>Our results indicated that EGCG improved cholesterol metabolism and meanwhile changed the lncRNAs expression profile in HepG2 cells. LncRNAs may play an important role in the cholesterol metabolism.</p>

ABSTRACT

Objective: To investigate the effects of epigallocatechingallate (EGCG) on lipid metabolism related gene and long non-coding RNA (lncRNA) expression proifle by biochip technology, and to explore the possible relationship between the two elements. Methods: HepG2 cell was cultured with EGCG at 25μmol/L for 24 hours, the total RNA was extracted and hybridized into the biochip of Human Transcriptome Array 2.0 for mRNA and lncRNA expression profile analysis. Bioinformatics technology was used to establish the possible relationship between lncRNA and the predicted target genes;the data obtained from biochip microarray was conifrmed by real time RT-PCR examination. Results: The microarray revealed that EGCG treated HepG2 cell expressed 27 differential lipid metabolism genes and 11 of them involved in cholesterol metabolism. In addition, there 285 lncRNA expressions were up-or down-regulated. Bioinformatics technology indicated that the predicted target genes for lipid metabolism might be cis-or trans-regulated by lncRNA;the data from real-time RT-PCR was consistent with the data from biochip microarray. Conclusion: Tea polyphenols improves lipid metabolism and lncRNA might be involved in the regulation of lipid metabolism related gene.
